چکیده
Abstract The interplay between T cells, dendritic cells and keratinocytes is crucial for the development maintenance of inflammation in psoriasis. GADD45 proteins mediate DNA repair different including keratinocytes. In immune system, GADD45a GADD45b regulate function activation both lymphocytes links epigenetic regulation through its demethylase activity. Here, we analyzed expression skin, circulating a cohort psoriasis patients their by inflammatory signals. Thirty (17 male/13 female) with plaque 15 controls subjects (7 male/8 female), were enrolled. Psoriasis exhibited lower at epidermis but higher dermal infiltrating lesional skin. was also peripheral from patients, although no differences observed p38 activation. methylation state target UCHL1 evaluated, revealing hypermethylation promoter skin compared to controls. Furthermore, reduced levels correlated We propose that may account pleiotropic effects, complex heterogeneous pattern psoriatic disease.

DNA damage and DNA damage-inducible protein Gadd45 following ischemia in the P7 neonatal rat.
Cerebral ischemia in adult rodents leads to the production of several types of lesions in the genomic DNA, followed by the activation of the damage-response indicator Gadd45. Our purpose was to investigate the structural changes that occur in chromatin DNA and repair processes after ischemic injury in neonatal brain. Neonatal ischemia was induced by the permanent left MCA occlusion in associati...
متن کاملRegulation of the Growth Arrest and DNA Damage-Inducible Gene 45 (GADD45) by Peroxisome Proliferator-Activated Receptor g in Vascular Smooth Muscle Cells
Peroxisome proliferator-activated receptor g (PPARg) is activated by thiazolidinediones (TZDs), widely used as insulin-sensitizing agents for the treatment of type 2 diabetes. TZDs have been shown to induce apoptosis in a variety of mammalian cells. In vascular smooth muscle cells (VSMCs), proliferation and apoptosis may be competing processes during the formation of restenotic and atherosclero...

متن کاملTransient global ischemia triggers expression of the DNA damage-inducible gene GADD45 in the rat brain.
Using in situ hybridization, Northern blot analysis, Western blot analysis, and immunocytochemistry, mRNA and protein expression of the novel DNA damage-inducible gene GADD45 was examined in the rat brain at 0.5, 2, 4, 8, 16, 24, 48, and 72 hours after 15 minutes of transient global ischemia. Transient ischemia produced by the four-vessel occlusion method resulted in DNA double-strand breaks an...
